Instructions for use of Artril in ampoules: description of the drug, its effectiveness in the treatment of joints

Arthril will help cope with acute joint pain

REFERENCE. The drug Arthril can be used for various diseases of the skeletal system and disorders associated with the normal functioning of the musculoskeletal system. This drug is indicated for rheumatoid arthritis, spondylitis, acute osteochondrosis, osteoarthritis of completely different localizations, polyarthritis and polyarthrosis, spondylosis.

If kidney function is impaired, this drug should be avoided.

In addition to side effects, this drug has its contraindications. It is not prescribed if the human body has an individual intolerance to one of the active components of this medicine. You cannot treat joints in this way in case of acute severe heart failure. If there are disturbances in the functioning of the kidneys and liver. Contraindications to this drug are:

  • pregnancy period;
  • breast-feeding;
  • children under 12 years of age;
  • exacerbation of stomach ulcers;
  • presence of internal bleeding.

ATTENTION! Arthril is also not recommended for use by those who have previously experienced allergic reactions after using non-steroidal drugs. And in order not to aggravate your condition due to joint disorders and not to cause even greater harm to yourself, such contraindications must be taken into account before starting to treat one of the diseases of the skeletal system.

With extreme caution, ampoules are prescribed to those who suffer from arterial hypotension. If a person is already taking sedatives, as well as drugs that help restore disturbed sleep, then it is extremely undesirable to use Arthril ampoules during such a period.

To get a truly positive therapeutic effect, it is necessary to properly organize therapy with such a drug. The instructions for use included with it will help with this. Arthril, released in ampoules, is used only for intramuscular administration. The duration of the entire course can be from four to six weeks. The drug is administered every other day.

In order to achieve maximum therapeutic results at home and cope with advanced joint inflammation, the combined use of this drug is recommended. Therefore, Arthril powder and ampoules should be used together whenever possible.

If side effects occur during the use of the medicine, and the person feels general malaise, then the recommended dosage of the drug should be reduced. It should be taken into account that for different pathologies of the skeletal system, the dosage is also different.

  • Instructions for use of Artril in ampoules: composition, analogues, reviews and price

When it comes to osteochondrosis in an acute stage, then the doctor can prescribe 3 ml of medication per day, once a day. With the development of spondylitis and rheumatoid arthritis, the medicine is prescribed in an increased volume. If the pain is too acute, Arthril ampoules are not recommended.

Instructions for use of Arthril are written in accessible language, which allows you to treat diseased joints at home effectively and efficiently. You can get rid of pain within an hour and a half after using this medicine. The inflammatory process begins to subside after a week of use.

Arthril in ampoules is administered only intramuscularly

The treatment regimen is quite simple. First, the active substance of the drug is mixed with the ampoule and placed in a syringe. The medicine is administered intramuscularly. The procedure is repeated three times a week. The duration of the entire course can be from four to six weeks. Over the entire treatment period, 12–18 ampoules may be needed.

Long-term use of Arthril injections can provoke the formation of blood clots. And if, due to the characteristics of the body, such a drug is not suitable for a person, you can try using its analogues that have a similar composition. These are Metipred, Rumalon, Artrocol, Artrosan. In any case, as soon as problems and pain in the joint area begin, it is necessary to immediately take action and begin treatment so that the disease does not take on an advanced form and does not provoke pathological processes in the bone tissue.

Diseases that affect the cartilage tissue of joints have similar names, etiology, symptoms and pathogenesis. Arthritis and arthrosis are pathologies that are partially similar and associated with damage to joint tissue, but classified as different classes of diseases.

Arthrosis is a lesion exclusively of the cartilage tissue of the joints, while arthritis is characterized by the development of inflammatory processes affecting, among other things, internal organs. The pathogenesis of pathologies also has a significant difference: arthrosis is often a consequence of age-related changes, while the causes of the development of arthritis are mainly damage to the body by infectious and viral agents. Differential diagnosis is a prerequisite, since making an incorrect diagnosis can lead to the choice of inappropriate treatment tactics and a decrease in the quality of treatment.

Features of application

"Arthril" in ampoules is prescribed with caution to patients with a history of chronic heart failure or arterial hypotension.

Drug interactions

"Arthril" in ampoules is prescribed very carefully in combination with the following drugs:

  • digitoxin;
  • amiodarone;
  • verapamil;
  • procainamide;
  • sodium thiopental;
  • polymiskin B;
  • cimetidine

It is worth noting that taking Arthril together with sedatives and medications for the treatment of sleep disorders, although possible, is undesirable.

Prescription during pregnancy and breastfeeding

Instructions for use note that clinical studies on the effect of glucosamine sulfate on the fetus and newborns have not been conducted. Since the drug contains lidocaine, it is prohibited for use by pregnant and lactating women.

Causes

The main difference between arthritis and arthrosis is the general characteristics of pathologies. Arthritis, regardless of the degree of development of the disease and its form, are inflammatory lesions of joint tissue, the occurrence of which can be caused by a wide range of negative factors. Arthrosis is characterized by degenerative-dystrophic changes that affect cartilage tissue due to age-related changes in the body or a number of other reasons.

  • Instructions for use of the drug Stop Arthritis (Super, Forte, Normal) for arthritis and arthrosis

Despite the significant differences, joint pathologies have similar pathogenesis and causes. The factors that provoke the development of arthrosis include the following:

  • Excessive physical activity.
  • General hypothermia of the body.
  • Violation of metabolic processes.
  • A large amount of excess body weight.
  • Age-related changes.
  • Leading a predominantly sedentary lifestyle.
  • Having bad habits.
  • Poor nutrition, consumption of large amounts of salt, fatty foods.

Arthritis can be caused by both identical causes and significantly different factors:

  • The patient's tendency to develop allergic reactions.
  • Metabolic disease.
  • Joint inflammation caused by physiological factors or exposure to viral agents.
  • Infectious diseases of other organs and systems. Infection of cartilage tissue and the development of arthrosis in such cases occurs through the systemic bloodstream.
  • Mechanical injuries.

As for the age trend, the development of arthrosis is diagnosed more often in older people. Arthritis, occurring in allergic or other forms, worries young people whose age does not exceed forty years.

Treatment methods

Treatment methods for arthritis and arthrosis are based on the use of medications, the action of which is aimed at suppressing pathogenic microflora, eliminating inflammatory processes, discomfort, pain and restoring cartilage tissue.

Most often, the list of drugs used in the treatment of joint pathologies includes the following:

  • Anti-inflammatory drugs. They are used to suppress inflammatory processes, eliminate discomfort and pain, and prevent the spread of infection through the systemic bloodstream.
  • Drugs that promote the restoration of cartilage tissue. They should be used in the treatment of arthrosis and arthritis, as they help restore damaged tissue and prevent further destruction of joints.
  • Drugs that stimulate metabolic and regenerative processes in tissues, as well as help saturate the body with important vitamins and microelements.

Complex treatment of joint diseases also includes the use of physiotherapy methods or other methods recommended by the attending physician. However, the prevailing and most effective method is the use of medications for arthritis and arthrosis.

Side effects

If the doses and frequency of administration are observed, side effects occur extremely rarely and are manifested by the occurrence of:

  • constipation;
  • diarrhea;
  • flatulence;
  • itching;
  • hives.

Since the drug contains lidocaine, this, in turn, leads to the following negative effects:

  • drowsiness;
  • diplopia;
  • dizziness;
  • headache;
  • tremor;
  • numbness in the mouth;
  • disorientation;
  • disturbance in the functioning of the heart.

pharmachologic effect

Mechanism of action Glucosamine sulfate is a salt of the aminomonosaccharide glucosamine, which is an endogenous component and the preferred substrate for the synthesis of glycosaminoglycans and proteoglycans of articular cartilage and synovial fluid. Glucosamine sulfate inhibits the activity of interleukin-1 beta and other inflammatory mediators. Clinical efficacy and tolerability The safety and effectiveness of glucosamine sulfate has been confirmed in clinical trials with treatment durations (oral administration) of up to three years. Short- and medium-term clinical studies have shown that the effectiveness of glucosamine sulfate in relation to the symptoms of osteoarthritis is observed after 2-3 weeks of its use. However, unlike NSAIDs, glucosamine sulfate has a long-lasting effect, lasting from 6 months to 3 years. Clinical studies with daily administration of glucosamine sulfate for up to 3 years have shown gradual improvement in symptoms and slowing of structural changes in the joint, as demonstrated by radiography. Glucosamine sulfate demonstrated good tolerability during short-term and long-term courses of treatment. Evidence of the drug's effectiveness was demonstrated when taken for three months, with residual effects for two months after its discontinuation. The safety and effectiveness of the drug have also been confirmed in clinical trials for up to three years. Continuous treatment for more than 3 years cannot be recommended, since there are no safety data when taking glucosamine for more than 3 years.

Pharmacokinetics

Glucosamine is a relatively small molecule (molecular weight 179) that is readily soluble in water and hydrophilic organic solvents. Available information on the pharmacokinetics of intramuscularly administered glucosamine is limited. Absolute bioavailability is unknown. The volume of distribution is approximately 5 liters and the half-life after intravenous administration is approximately 2 hours. About 38% of an intravenously administered dose is excreted unchanged in the urine.

Indications for use

Relief of symptoms (mild to moderate pain) of adequately diagnosed osteoarthritis of the knee.

Directions for use and doses

For intramuscular use! The medicine is not intended for intravenous administration. Before use, ampoules with the drug and solvent are kept until they reach room temperature in order to prevent discomfort at the injection site. When initiating a course of treatment, the drug Artrolek can be stored at room temperature in its original packaging for no more than 6 months. Adult and elderly patients. Before use, mix solution B (solvent 1 ml) with solution A (drug solution 2 ml) in one syringe. The prepared drug solution is administered intramuscularly in 3 ml or 6 ml (solution A+B) 3 times a week for 4–6 weeks. The presence of a yellowish color of the solution in ampoule A does not affect the effectiveness and tolerability of the drug. Intramuscular use can be combined with oral administration of the drug Artrolek in powder form to prepare a solution. Glucosamine is not intended for the treatment of acute pain. Relief of symptoms (especially pain) may only occur after several weeks of treatment, and in some cases even after longer. If no relief of symptoms has occurred after 2–3 months of use, it is necessary to reconsider (the advisability of continuing) treatment. Patients should consult a doctor if pain worsens after starting glucosamine. Dosage regimen for different categories of patients Elderly patients No dose adjustment is required. Patients with impaired renal and/or liver function In patients with impaired renal and/or liver function, no recommendations for dose adjustment exist, because no relevant studies have been conducted. Children and adolescents Glucosamine should not be used in children and adolescents under the age of 18 years, since there is no data on the safety and effectiveness of glucosamine in this category of patients.

Use during pregnancy and lactation

The injection form of the drug Artrolek includes the excipient lidocaine, which has the following contraindications: arrhythmia, acute heart failure, and hypersensitivity to lidocaine.

Precautionary measures

It is necessary to consult a doctor to rule out the presence of joint diseases for which other treatment methods are provided. Cases of exacerbation of bronchial asthma symptoms have been described after starting to take glucosamine. Patients suffering from bronchial asthma should be informed about the possible worsening of symptoms of the disease. One dose of the drug contains 40.3 mg (1.75 mmol) sodium. This should be taken into account when prescribing to patients on a sodium-restricted diet. Patients with impaired glucose tolerance should use caution when taking glucosamine. For patients with diabetes mellitus, it is recommended that blood sugar levels be monitored and insulin requirements determined before and periodically during treatment. No specific studies have been conducted in patients with renal or hepatic impairment. The toxicological and pharmacokinetic profiles of glucosamine do not suggest restrictions for these patients. However, the use of glucosamine in patients with severe hepatic or renal impairment should be under medical supervision. Use during pregnancy and breastfeeding There are no data on the use of the drug in pregnant women or during breastfeeding, so use of the drug during pregnancy or breastfeeding is not recommended. Effect on the ability to drive and operate machinery No special studies have been conducted on the effect of glucosamine on the ability to drive and operate machinery. However, if headaches, drowsiness, fatigue, dizziness or visual disturbances occur while taking glucosamine, driving or operating machinery is not recommended.

Interaction with other drugs

No specific drug interaction studies have been conducted. There is evidence of an enhanced effect of coumarin anticoagulants; therefore, more careful monitoring of coagulation parameters is necessary in patients simultaneously taking coumarin anticoagulants (for example, warfarin or acenocoumarol). Oral administration of glucosamine sulfate may increase the absorption of tetracyclines from the gastrointestinal tract, but the clinical significance of this interaction is small. It is acceptable to take steroidal or non-steroidal anti-inflammatory drugs simultaneously with glucosamine.

Contraindications

Individual hypersensitivity to the active substance or to any of the excipients. The drug should not be used by patients with allergies to shellfish, since the active substance (glucosamine) is obtained from the shells of mollusks and crustaceans.

Compound

Ampoule A (solution A) Active ingredient: glucosamine sulfate (in the form of glucosamine sulfate sodium chloride) – 400 mg (502.5 mg). Excipients: lidocaine hydrochloride, 1M hydrochloric acid solution or 1M sodium hydroxide solution, water for injection. Ampoule B (solution B (solvent)) Excipients: diethanolamine, water for injection.

Overdose

Cases of overdose are unknown. In cases of overdose, symptomatic treatment should be carried out aimed at restoring water and electrolyte balance.

Side effect

General profile of adverse events: The most common adverse reactions associated with intramuscular administration of glucosamine are nausea, abdominal pain, dyspepsia, flatulence, constipation and diarrhea, and rarely skin irritation, itching and redness. Injectable forms of glucosamine may cause nausea and vomiting (rarely) due to the lidocaine content. These adverse reactions, as a rule, were moderate and transient. The following adverse reactions have been grouped based on the MedDRA classification. When using glucosamine, undesirable effects are registered with the following frequency: very often - 1/10 or more; often - from 1/100 to 1/10; infrequently – from 1/1000 to 1/100; rarely - from 1/10,000 to 1/1000; very rare - less than 1/10,000, frequency unknown - cannot be determined based on available data. From the immune system: frequency unknown - allergic reactions*. From the side of metabolism and nutrition: frequency unknown - inadequate glycemic control in diabetes. Mental disorders: frequency unknown - insomnia. From the nervous system: often – headache, drowsiness; frequency unknown - dizziness. On the part of the organ of vision: frequency unknown - visual impairment. From the heart: frequency unknown - arrhythmias, including tachycardia. From the vascular system: infrequently - hot flashes. From the respiratory system, chest and mediastinum: frequency unknown - asthma / worsening of asthma. From the gastrointestinal tract: often – diarrhea, constipation, nausea, flatulence, abdominal pain, dyspepsia; frequency unknown - vomiting. From the skin, subcutaneous tissue: infrequently - erythema, itching, rash; frequency unknown - angioedema, urticaria. From the liver and biliary tract: frequency unknown - increased levels of “liver” enzymes in the blood and jaundice**. General disorders: often – fatigue; frequency unknown - edema/peripheral edema, reactions at the injection site. From laboratory and physiological indicators: frequency unknown - increased liver enzymes, blood glucose levels, increased blood pressure, fluctuations in MHO levels. * Predisposed patients may develop severe allergic reactions to glucosamine. **Cases of elevated liver enzymes and the development of jaundice have been reported, but a cause-and-effect relationship with glucosamine intake has not been established. Cases of hypercholesterolemia have been reported, but a causal relationship with glucosamine intake has not been demonstrated.

Storage conditions

Pharmacological features of the drug

Let us consider in detail the pharmacodynamics and pharmacokinetics of Artril.

Pharmacodynamics

Glucosamine stimulates increased formation of proteoglycans, which are found in cartilage fluid, and stops the production of enzymes that destroy cartilage. Main effects of glucosamine:

  • sulfur participates in the formation of chondroitinsulfuric acid;
  • calcium accumulates in sufficient quantities in the bones;
  • the optimal level of glycosaminoglycans is restored;
  • the negative effect of corticosteroids on chondrocytes is reduced.

The medicine restores the amount of glucosamine, has a beneficial effect on capsular permeability and accelerates the restoration of normal enzymatic processes in cartilage cells.

Pharmacokinetics

The drug is quickly and almost completely absorbed from the small intestine. After intramuscular administration, it reaches the joints within a short time. The half-life of the drug is about three days.

The patient begins to feel the therapeutic effect of the drug after 10-14 days of systematic use. The results of using the medicine last for about two months after completion of the course.

Patients noted the following positive phenomena in the body after using the pharmaceutical product:

  • minimizing pain;
  • improving joint mobility;
  • absence of crackling and crunching;
  • There is no longer any stiffness, numbness or swelling in the limbs.

Pharmacodynamics

The active substance of the drug "Arthril" undergoes complete and rapid absorption from the small intestine. After administration intramuscularly or orally, it easily enters the articular cartilage. The half-life is quite long and is almost 3 days (68 hours). The patient can feel the first improvements after two weeks of drug therapy, and the achieved positive dynamics last for about 8 weeks after the course.

As a result of treatment, patients note the following results:

  • pain decreases;
  • joint mobility increases;
  • the crackling and crunching disappear;
  • the feeling of stiffness, numbness and swelling disappears.

Indications and contraindications

The official instructions for use state that the medicine is prescribed to patients who suffer from:

  • osteoarthritis of various locations (knee, hip, ankle, elbow and wrist);
  • arthritis and polyarthritis;
  • arthrosis and polyarthrosis;
  • coxarthrosis;
  • spondylosis;
  • spondylitis;
  • humeroscapular periarthritis;
  • chondromalacia patella.

  • procainamide;
  • sodium thiopental;
  • polymyxin B;
  • cimetidine

Concomitant use with sedatives and sleeping pills is possible, but undesirable.

Prescription during pregnancy and breastfeeding

Instructions for use note that clinical studies on the effect of glucosamine sulfate on the fetus and newborns have not been conducted. Since the drug contains lidocaine, it is prohibited for use by pregnant and lactating women.

Side effects

Experts say that if the patient strictly follows the instructions for use, Arthril does not cause negative effects. However, some people noted that while using the powder they began to worry:

  • constipation or diarrhea;
  • increased gas formation;
  • itching or hives.

Since one of the components of the drug is lidocaine, in some people it can provoke the development of:

  • drowsiness;
  • diplopia;
  • vertigo;
  • headaches;
  • trembling limbs;
  • disorientation;
  • disturbances in cardiac functionality.

Instructions for use

The drug "Arthril" in ampoules is used exclusively for intramuscular use. The course of treatment is from 4 to 6 weeks with intervals of taking 1 dose of the drug every other day.

The instructions describe the following rules for using the drug: before administering the medicine, you must mix the contents of the ampoule with the active ingredient (A) and the solvent (B). To achieve maximum effect, the combined use of Arthril in ampoules and bags is recommended.
